The College of Doncellas Nobles was founded in 1551 by the Archbishop of Toledo Cardinal Silíceo and, as per his wishes, the college has been co-sponsored by the King and the Archbishop of Toledo ever since.

Although the relief by Juan Bautista Vázquez the Elder on the main facade has survived from the original college, the current building was erected in the 18th century, in part by José Hernández Sierra in 1757-68 and then by Ventura Rodríguez in 1775-87. The college became so popular at the end of the 19th century that the site was extended with an adjoining building erected in 1900-1903 by Santiago Castellanos, owned by the Castile-La Mancha government since 1990.
